The finely minced tissue is extracted with cold trichloro-acetic acid to remove acid-soluble P and with ethyl alcohol-ether and methyl alcoholchloroform to remove lipoid P. The residue is digested withNpotassium hydroxide for at least 15 hr. at 37° C. to break down ribonucleic acid into its constituent nuoleotides and to split inorganic P from phosphoproteins. Total P is estimated in a portion of the alkaline digest. This gives a measure of ribonucleic acid P plus desoxyribonucleic acid P plus phosphoprotein P. Another portion of the alkaline digest is treated with HC1 and trichloroacetic acid to precipitate desoxyribonucleic acid. Total P and inorganic P are determined in the supernatant fluid. The difference between them gives the amount of ribonucleic acid P, while the inorganic P is a measure of the phosphoprotein P. The desoxyribonucleic acid P is given by the difference between the total P in the alkaline digest and the total P in the supernatant fluid after acidification of the digest.-J. N. Davidson.
