METHODSDNA constructions. The vector plasmids for transformation were pRPLI (Garabedian et al. 1985) and pCP20. 1 (Simon et al. 1985). The yp genes in the series ofypl 5'deletions were marked by insertion of the 676-bp M13mp8 BglII/XmnI fragment containing gene II (van Wezenbeek et al. 1980; Messing et al. 1981). It was inserted (Garabedian et al. 1985) into the XhoI site of ypl (Hung and Wensink 1981) and replaced a 393-bp fragment between the Stu [and BglII sites of yp2 (Hung and Wensink 1983). The resulting constructions were inserted into pRPLI. The series ofypl 3'deletions was constructed using two plasmids that contain the hsp70-lacZ fusion gene (Lis et al. 1983; Costlow et al. 1985). These plasmids were generously given to us by John Lis (Cornell University) and his colleagues. One of them has a XhoI site 196 bp upstream of the hsp70 cap site, the other (a deletion derivative of the first) has an XhoI site 43 bp upstream of the hsp70 cap site. We placed XhoI linkers at different locations upstream of the ypl promoter and then joined upstream regions of ypl to the appropriate hsp70-lacZ fusion via the XhoI linkers. The ypl-hsp70-lacZ fusion genes were inserted into pCP20. 1 (Figs. 1 and 5, below). The methods of construction are described elsewhere (Schleif and Wensink 1981; Maniatis et al. 1982; Garabedian et al. 1985).Germ line transformation. The method of Spradling and Rubin (1982) was used to transform ry-embryos (ry~ a gift from Welcome Bender, Harvard University Medical School) with DNA constructs that included a ry § gene. Adults derived from injected embryos were mated to ry-flies. The ry+ progeny were selected and mated to ry-flies. The ry+ progeny of this mating were parents of the resulting transformed line.
