The membrane potential of frog muscle fibers varied in the same manner as a potassium electrode when [K]o and [Cl]0 were changed reciprocally at constant product, or if [K]o was altered in Cl-free solutions. At constant [K]o, variation of [Cl]0 produced no permanent displacement of membrane potential, but there were large transient changes. At constant [Cl]0, increasing [K]o from 2.5 to 10 mM produced a sudden depolarization and then a slow drift to an equilibrium value. Varying the internal chloride concentration, by altering the external osmotic pressure with sucrose, changed the membrane potential of a fiber loaded with Cl in the manner expected for a chloride electrode. These and additional data are explained on the basis of assuming a Donnan system of the Boyle-Conway type.
