The first recognition that there was a familial relationship between the serine proteinase inhibitors antithrombin and acproteinase inhibitor (also known as a 1-antitrypsin) was made in 1980 by Hunt and Dayhoff (2) on the basis of comparison of primary structure information for these inhibitors as well as the non-inhibitory protein ovalbumin. It is now known from classical and DNA-derived protein sequence data on more than 60 proteins that this familial relationship extends to proteins from sources as diverse as viruses and humans and from a variety of locations from plasma to neural glia (3). From X-ray structure determinations on several members it is now realized that the primary structural similarities extend to tertiary structure similarities, with a core domain of about 350 amino acids being common to each member of the family (4-13). The acronym coined by Carrell and Travis (14) to describe members of the family is SERPIN, standing for serine nroteinase inhibitor, although certain serpins, notably ovalbumin, thyroxine binding globulin, and corticosteroid binding globulin, are not known to be inhibitors of serine, or other proteinases. One of the main reasons for interest in serpins is that a number of them play critical roles in the regulation of important physiological processes such as blood coagulation (e. g. antithrombin), fibrinolysis (e. g. plasminogen activator inhibitor 1), complement activation (Cl inhibitor), and inflammation (acproteinase inhibitor).
