Identification of prostaglandins E1 E2 F1X and F2X in the amniotic fluid is the subject of this report. Amniotic fluid was obtained from 20 patients at term following spontaneous rupture of the fetal membranes in normal pregnancies. The silicic acid chromatographic procedure used gave 4 peaks of biological activity. 2 of these peaks had the chromatographic and biological properties expected of prostaglandins Es and Fs respectively. With the aid of tritium labelled PGE1 and PGF1x Samulesson (1963) showed that prostaglandin Es can be eluted with ethyl acetate: benzene (60:40) and those of F series with ethyl acetate -- benzene (80:20) from silicic acid column. The biologically active material from peaks 2 and 3 of silicic acid chromatography were further examined for the presence of prostaglandin Es and Fs respectively. The identification was achieved by thin layer chromatographic procedure as described by Green and Samuelsson (1964). The material from fractions 5-11 of silicic acid column had chromatographic properties identical to those of prostaglandins E1 and E2 when compared with pure prostaglandins either as free acids or methylesters on 3 different solvent systems. The Rf values of material from fractions 12-17 were identical to the Rf values of free acids or methyl ester of prostaglandins F1X and F2X.
