Since the discovery of ET-1 in 1988 [1], few molecules have come under such intense scrutiny as the endothelin peptide family (ET-1, ET-2, ET-3) and their receptors (ETA, ETB, ETC). Much study had nevertheless given rise to, at best, a limited understanding of their regulation and roles in health and disease. Perhaps it was not surprising that'endothelin fatigue'had begun to sink in, even among the most committed researchers!In the past year, however, several new and significant developments have led to a renewed interest in these peptides. Firstly endothelins and their receptors have been shown to be essential for the development of fetal neural crest derived tissues in mice. In addition, they have been implicated in at least two specific human diseases [2-6]. Secondly the endothelin converting enzyme (ECE-1), an important therapeutic target for modulating endothelin synthesis and action, has been cloned [7]. Thirdly selective, potent and orally active endothelin receptor antagonists are now available [8-10]. Finally an endothelin receptor antagonist given 24 h after renal ischaemia, has been shown to reverse severe acute tubular necrosis in rats, by specifically stimulating tubular recovery [11]. Thus, although the major focus in endothelin research has been on the regulation of vascular tone (because of the nature of their discovery), it is now apparent that some of the essential functions of the endothelins are actually on growth and development in non-vascular tissues. For a more comprehensive account of the roles of endothelins in the kidney, the interested reader may refer. to several recent accounts [12, 13]. It is not my purpose to add to this literature. Rather, since the major focus of endothelin renal research has inevitably concentrated on the control of glomerular function and vascular tone, my aim is to show that the tubulointerstitium is in fact, the major site of ET-1 synthesis and action in the kidney.
