Herpes simplex virus particles of a strain which causes syncytium formation (macroplaques) and a strain which causes rounding and clumping of cells (microplaques) were ultracentrifuged in a solution of cesium chloride. Assays of infectious macroplaque and microplaque virus particles contained in fractions collected from the bottom of the tube after the ultracentrifugation revealed that the two strains differ in their apparent buoyant density by 0.8%. The macroplaque virus which localized in less dense zones may be less homogeneous with respect to density than the microplaque variant of herpes simplex virus. It is concluded that the physical difference between the two strains may be due either to a difference in nucleic acid, protein, or lipid content of the particles or to preferential interaction of particles of one strain with cesium chloride or impurities
