Method of mapping: Cox8h was localized by haplotype analysis of 94 progeny from an interspecific backcross,(C57BL/6J x Mus spretus) F 1 x Mus spretus from The Jackson Laboratory [1]. Database deposit information: The scorings are available on the internet at the following address: http://www. jax. org/crossdata. html.Molecular reagents: As a hybridization probe, we used the 1142-bp intron of the mouse Cox8h gene. The intron was isolated by PCR from a mouse Cox8h genomic clone from a strain 129/SVJ genomice library (Stratagene, LaJolla, CA)[M. Lomax, unpublished data] with primers designed to the cDNA sequence [2]. Exon 1 primer: 5'TGTGCAACTTCATGCTGC 3'; exon 2 primer: 5'GAAGTTCACAGTGGTTCC 3'. PCR (100 Ixl final volume) conditions were as follows: 0.6 ng/txl template, 1 x PCR buffer (Gibco-BRL, Gaithersburg, MD), 0.75 mM MgC12, 100 tXM each dNTP, 1 IXM each primer, and 0.025 units/l~ l Taq DNA polymerase (Gibco-BRL). Thermal cycling was performed for 30 cycles of 94~ for 45 s, 55~ for 45 s, and 72~ for 1.5 min. The reaction resulted in a single, 1142-bp product that was subjected to electrophoresis through an agarose gel. The DNA was eluted from the excised gel band with an Ultrafree-MC 0.45~ m filter (Millipore) and labeled with [a-32p] dCTP (> 3000 Ci/mm; Dupont-NEN) by the random primer method [3].
