Primordial germ cells were found to possess such a high degree of alkaline phosphatase activity as demonstrated by the . revised method of Gomori (1946) that it was possible with short incubation periods to stain the cells almost selectively and to trace their migration from the yolk sac to the germinal ridge. Distribution of germ cells was studied in. serial sections of embryos from 6 to 12 days'' copulation age. Germ cells were observed on the 8th day, scattered among the cells of the caudal end of the primitive streak, the root of the allantoic mesoderm, and the adjacent yolk sac. It is believed that germ cells probably originate in the yolk sac splanchnopleure. Later migration of the cells in 9- to 12_day embryos is along the open mid gut and stalk of the allantois to the hind gut and then dorsally within the dorsal mesentery to attain their final position in the germinal ridges. Significance of the high enzymatic activity of the germ cells is not known but it is not characteristic of all migrating embryonic cells.
