为进一步了解戊型肝炎病毒(HEV)的感染机制和致病机理,我们从HepG 2细胞中提取HEV病毒样颗粒(p239),通过亲和层析、双向电泳和质谱技术筛选与p239相互作用的蛋白,并筛选出GRP 78/Bip、HSP 90、通过免疫共沉淀法进一步证实了GRP 78/Bip与p239的相互作用。Bip可能有助于HEV的细胞粘附或颗粒组装。
For further understanding the infection mechanism and pathogenesis of hepatitis E virus(HEV),we screened the protein interacting with HEV virus-like particle(p239) extracted from HepG2 cells by affinity chromatography,two-dimensional electrophoresis and mass spectrometry.Four candidate proteins,GRP78/Bip, HSP90,alpha-tubulin and P43 were identified.The interaction between GRP78/Bip and p239 was further confirmed by co-immunoprecipitation.Our results suggest that GRP78/Bip may contribute to the cell attachment or particle assembly of HEV.
