A serial electron microscope study was performed on developing fungiform papillae and taste buds from tongues of rats ranging in age from the 16th day of gestation to 28 days postnatally. For convenience of description the developmental process was arbitrarily divided into 5 stages. Fungiform papillae, containing both epithelial and connective tissue components, appear first on the 16th day of gestation (stage 1). This is followed by the appearance of nerve fibers within the connective tissue core (stage 2) and then the entrance of nerve processes into the epithelium at the free surface (stage 3). The latter is associated with the striking appearance of clusters of vesicles in the epithelial cells adjacent to nerve processes. Several days later, the cells in the forming taste bud have begun to differentiate into 3 different types (stage 4); finally, a 4th cell type appears at about the time that the taste pore is established (stage 5).
