A radiometric determination of PPi is described. Excess UDP[14C]glucose is used to react with PPi in a reaction catalyzed by UDP-glucose pyrophosphorylase. This reaction liberates [14C]glucose 1-phosphate, which is then further converted to 6-phosphol[14C]gluconic acid in the presence of phosphoglucose mutase and G-6-P dehydrogenase. The reaction product, 6-phosphol[14C]gluconic acid, is separated from UDP-[14C]glucose by the addition of activated charcoal, which adsorbs UDP-[14C]glucose preferentially. The sensitivity of the assay for PPi is 20 pmol.
