The nuclear DNA complement of Hemophilus influenzae has been determined to be 700 to 800 X 106 daltons. An isolation procedure has been developed which gives high yields of DNA with a maximum molecular weight of 400 X 106, or one-half of the nuclear DNA complement. Linkage of previously unlinked markers has been obtained in transformation assays. The isolation procedure has been extended to Escherichia coli and Serratia marcescens.
