By means of the methods of Druet, May, Henderson, Packman, and Peacock, the site and degree of retention of inhaled particles were studied. The spores of Bacillus subtilis were used. The spores were made radioactive by growing B. subtilis in a medium containing P32 as Na2HPO4. Aerosols of the labelled spores were then made with the Druett and May apparatus. Guinea pigs, monkeys, and mice were exposed to the aerosols and were killed and examined either Immediately after exposure or up to 24 hrs. after exposure. The head, trachea, lungs, stomach, and gut were examined for the spores. They observed that the proportion of spores retained in the head increased with increased particle size. With particles of a diam. of 1 [mu], only 3-7% of the spores were retained, whereas with particles having a diameter of 6[mu], 75-85% were retained in the head. The trachea never retained the spores in great nos.. When the spores left the head, they passed directly to the deep recesses of the lungs or to the stomach; from there they were rather quickly removed from the body.
