The acidic soluble proteins of chromaf6n granules have been collectively named chromogranins.'During the last 20 years these chromogranins have been analyzed and characterized in great detail (for review see refs. 2-5), their physicochemical properties and biosynthetic pathways established, and in 1986 the primary sequence of one chromogranin was obtained for the first time by Iacangelo et aL6 Nonetheless, the physiological function of these characteristic proteins is still unknown. Originally it was thought that these proteins are involved in a storage complex with catecholamines. More recent studies demonstrating a widespread distribution of these proteins in several nonadrenergic endocrine and neuronal tissues have, however, called into question this finding. In this paper we summarize recent evidence demonstrating similarities of the chromogranins with neuropeptides and regulatory peptides which suggest that these proteins might have a function after secretion into circulation.
