HEART EXCITATORY AND INHIBITORY SUBSTANCES IN MOLLUSCAN GANGLIA

Nerve and heart tissue extracts from some molluscs are known to contain heart excitatory material in addition to 5HT. We have used Sephadex G-15 columns to analyze this material. Eluted fractions were assayed on hearts of the clam Mercenaria mercenaria treated with 1-methyl-lysergic acid butanolamide and with Mytelase in order to counteract 5-hydroxytryptamine and acetylcholine. Extracts from ganglia of M. mercenaria gave rise to 4 peaks of cardioexcitatory activity (peaks A, B, C, D). Extracts from clam hearts gave rise only to peaks A, B and C. Extracts from ganglia of the snail Busycon canaliculatum gave rise to peaks C and D. Peaks A, B and C from either the clam or the snail were completely inactivated by treatment with proteolytic enzymes; peak D was not inactivated.
