The history of in-cell NMR spectroscopy started as the high-resolution multidimensional NMR spectroscopy of isotope-labelled proteins in Escherichia coli cells1 because of well-established sample preparation methods such as selective overexpression and stable isotope labelling of the target proteins. Indeed, various applications utilizing E. coli cells were proposed after the first report of in-cell NMR experiments. 2–7 However, the limited employment of cell types has only limited applications in terms of ‘‘in situ’’observation of biological macromolecules especially proteins, meaning that only E. coli proteins should be investigated in E. coli cells. Unlike the case of
