NUCLEAR UPTAKE OF DNA FRAGMENTS AFTER INJECTION OF P-32-LABELLED THYMOCYTES IN LETHALLY IRRADIATED MICE

Mice, total-body X-irradiated with 800 r 24 hr previously, were injected intravenously with homologous p32-iabelled thymocytes, either living or killed by UV irradiation. The control mice were given inorganic P32 of the same activity as that of the cell suspension, i. e. 0.25 [mu]C per mouse. The organ distribution of radioactivity was followed in the homogenates of the thymus, spleen, lungs, liver, kidneys, and duodenum, and by high resolution autoradiography. There was greater retention of radioactivity at 24 and 48 hr after the injection of cells than after inorganic P32 in the spleen and liver. Labelled thymocytes were found in the thymus, spleen and bone marrow. After the injection of labelled cells as well as inorganic p32; the sinusoids, cytoplasm, and nuclei of liver cells were labelled in the liver; the glomerular capillaries, Bowman''s capsule, and both the proximal and distal tubules in the kidneys; and the epithelium and mucous membrane in the duodenum. Diffuse radioactivity was found in the thymus, spleen, and bone marrow autoradiographs. After the removal of non-DNA phosphorus by Feulgen hydrolysis in specimens of mice injected with p32- thymocytes, the residual radioactivity was localized in Feulgen positive fragments, lymphocytes, nuclei and cytoplasm of reticular cells, and in neutrophils of the thymus, spleen and bone marrow and, furthermore, in the endosteal cells of the femur. It is concluded that the injected thymocytes disintegrate in the lymphoid organs; the large fragments of their DNA are taken up by the cytoplasm and incorporated into the nuclei of the non-dividing reticular and endosteal cells.
