THE EFFECT OF CHEMICAL AND PHYSICAL AGENTS ON THE PHAGE RECEPTOR OF PHASE-II SHIGELLA-SONNEI

Because different prepns. of the somatic antigen obtained from Phase II S. sonnei exhibit variations in their ability to inhibit T3, T4 and T7, coli-dysentery phages a study was made of the effects of physical and chemical agents on the phage-adsorption properties of intact bacterial cells. A comparison of the virus adsorption rates on live and dead bacteria served as the criterion for the destruction of phage receptors. If the adsorption rates were equal it was assumed that no alteration of the receptor occurred following treatment of the microorganisms. The bactericidal agents studied included phenol, formol, ethylene oxide, ethanol, thymol, Na periodate, chloroform, heat, u.-v. irradiation, osmotic shock, sonic vibration and freezing and thawing. It was found that when Phase II S. sonnei was treated by any of the above procedures the rate of adsorption of T7 to the dead organisms was little altered, whereas the rate of T3 adsorption was considerably reduced. The adsorption rate of T4 was in some cases depressed but to a relatively lesser degree than that of T3. Only mild irradiation of the bacteria with u.-v. permitted the T3 virus to adsorb to the dead cells at a rapid rate. In all cases the 3 viruses adsorbed to the treated microorganisms at rates which followed the order T7>T4>T3. A detn. of the latent periods of the 3 viruses in Phase II S. sonnei gave values of 17 [plus or minus] 0.5 min. for T3, 24.5 [plus or minus] 0.5 min. for T4 and 20 [plus or minus] 0.5 min. for T7 when nutrient broth served as the culture medium.
