Acentral problem in cell biology is cellular movement and locomotion. Many workers investigate the locomotion of amoebre and several mechanisms have been proposed to account for the phenomena observed1-3• We have come to the conclusion that a satisfactory account of amoeboid locomotion must accommodate observations first made by Dellinger• many years ago. It has become clear during our work on the control of amoeboid locomotion that to examine an amoebre in the usual way under a coverslip, in a capillary tube, or a shallow glass chamber gives a very incomplete picture of the situation. We have re-investigated the sequence of events in locomotion using Dellinger's method of observing amoebre from the side and without a coverslip near to the amoebre. In general, Dellinger's description is confirmed and our work has revealed additional facts. We have used Amoeba proteus starved for 24 h, washed and observed in Chalkley's solution on clean glass. Amoeba dubia (King's College strain) and Pelomyxa carolinensis were also examined. The amoebre were observed either in a glass chamber,(Fig. 1), which allowed observation from the side and the top, or in a chamber with a prism which allowed a side view of the amoebre,(Fig. 2).
