Investi-gations were carried out on the nutrient requirements of suspension cultures derived from root sections of soybean (Glycine max. L.), and a simple medium was developed. The medium consists of mineral salts, sucrose, the B-vitamins, m-inositol and 2,4-dichlorophenoxy-acetic acid (2, 4-D). An absolute requirement for thlamine and 2, 4-D was demonstrated. N was supplied as nitrate and ammonium. Ammonium or some other source of reduced N was necessary for growth. Optimum concentration of KNO3 and (NH4)2SO4 were 25 and 1 mM, respectively. The highest yield of cells was achieved at an initial pH of 5.0. During the growth cycle the pH gradually increased to 6.2.
