Five years ago I published “Hypothesis: Calcium Modulated Proteins Contain EF-Hands.”’ The list of homologs now includes S-100, intestinal calcium-binding protein, parvalbumin, regulatory and essential light-chains of myosin, troponin C, and most important, calmodulin. A calcium-modulated protein is defined by its presence in the cell cytosol or on a membrane facing the cytosol and by its calcium affinity, pK, (Ca” ) 5 to 7 under cytosolic conditions. None of the calcium modulated proteins, characterized to date, are themselves enzymes. Yet by their interactions they confer calcium modulation on a wide variety of cytosolic enzymes and cell processes.
