CHANGES IN MARROW RESERVE OF EOSINOPHILS FOLLOWING RE-EXPOSURE TO FOREIGN PROTEIN

Quantitative methods were used to study the discharge of eosinophils from the bone marrow, following the re-exposure of sensitized guinea-pigs to foreign protein. A striking circulatory eosinophilia was observed in a group of guinea-pigs 29 hours after the last of four subcutaneous injections of foreign protein at 5 day intervals. As compared with a group in which the fourth injection was replaced by saline, the total number of marrow eosinophils showed a marked decrease, which was almost entirely the result of a decrease in the marrow "reserve" of mature and nearly mature eosinophils. No significant change was seen in the numbers of earlier forms. It appears that in the 29 hours following the reinjection of antigen, very large numbers of band and segmented eosinophils had been discharged from the marrow reserve into the blood stream. Calculation of the total blood and bone-marrow populations indicated that less than 10% of the discharged eosinophils were still present in the circulation. The findings indicate the importance of the marrow reserve of mature and nearly mature eosinophils to the defense mechanisms of the body. It enables the bone marrow to meet urgent demands for new eosinophils without the delay which would be inevitable if their supply depended on new production. Incidental observations showed rather similar changes in the basophil series, but no changes were observed in the neutrophil series nor in any other cell type.
