在这项研究中,一种新的聚合酶链反应(PCR)为基础的基因分型方法,以确定在日本生长的三个Corbiculasspecies(Corbicula japonica,Corbicula sandai,Corbicula laena)的发展。
In this study, a novel polymerase chain reaction (PCR)-based genotyping method to identify threeCorbiculaspecies growing in Japan (Corbicula japonica, Corbicula sandai, andCorbicula laena)was developed. Using an allele-specific PCR method,C. japonicacould be identified and specifically distinguished from the speciesC. sandaiandC. laenain both tissues and various processed foods. In addition, the discrimination ofC. sandaiandC. laenawas possible using the PCR-restriction fragment length polymorphism (RFLP) method withNlaIII as a restriction enzyme. Moreover, a preliminary experiment to determine the district in whichC. japonicagrows from among seven areas was performed. The use of the PCR-RFLP method made it possible to identifyC. japonicafrom Lake Ogawara. By using electrophoretic genotyping methods in the present study,C. japonicaspecies could be identified and the species of a particular growing district could be specified easily and reproducibly. This method is useful for protecting bioresources and maintaining ecosystems. The present study represents a contribution to the field because the strategy presented here can be applied to the forensic identification of other species, such as other bivalve and Mollusca.
