Histochemical studies of the binding of Congo red by amyloid and comparison of the staining of amyloid with various sulfonated dyes demonstrate that the selective staining of amyloid with Congo red can not be explained by interaction between sulfonic acid groups of the dye and basic groups of amyloid. The selective staining with direct cotton dyes, but not with other disazo dyes, indicates correlation between the distribution of hydrogen bonding groups in the dye molecule and affinity for amyloid. It is suggested that the selective staining of amyloid with Congo red is due to steric configurations of amyloid which favor non-ionic binding of the dye, probably by a mechanism similar to that operative in the dyeing of cellulosic materials.
