Pieces of human skin obtained at operation or by biopsy from subjects of both sexes aged 3-70 years were examined. Specimens were taken from abdomen, shoulder, breast, loin, calf and foot. Material was fixed by a variety of methods including buffered formol-dichromate (pH6). The sections were prepared and stained by a variety of methods. Three types of human mast cells were identified. Melanin was localized in both epidermal and dermal cells. Some mast cells and dermal melanophores are polymorphic and possess attenuated branching processes. These cells have an affinity for Giemsa stain. The staining reactions of adrenal medullary and para-aortic chromaffin cells were compared with those of the granular cells of skin. No evidence of pressor amine-containing dermal chromaffin cells was found.
