MATERIALS AND METHODSThree metals were chosen for this experiment: cobalt-chromium alloy (ASTM F75-76), 6 Multiphase'35% Nickel (ASRVI F% 2-78),'and commemdy pure titanium (ASTM F67-77).* Adult mongrel dogs were implanted with solid specimens 8.9 mm in length by 4.4 mm in diameter. One specimen was placed in the supracondylar region and a second specimen was placed in the diaphyseal region of the femur, perpendicular to the long axis of the bone. This procedure was done bilaterally in nine dogs, three dogs-metal tested.One experimental time period of 2 years was chosen. After sacrifice, the implants with the surrounding bone were retrieved. They were fixed in 6% gluteraldehyde for 12 h, then rinsed and embedded in expoxy resin. The sample block was then cut in half and the surface polished to 1.0 p~ with diamond. Sections both with the implant and with the implant removed were carbon coated to make them electrically conductive for electron microprobe analysis. An ARL EMX-SM microprobe analyzer with three crystal spectrometers was used. Microprobe data as counts versus distance was collected at several areas in each sample to determine the concentration gradient of metal (titanium, nickel, cobalt, or chromium) in the vicinity of the implant. All four metals in the pure form were used as a standard with appropriate correction for a lower density medium. Additionally caldumphosphorus ratios were determined to venfy the sampling area was in fact mature bone.
