Isolates of Haemophilus influenzae type b may be subtyped by differences in the patterns of their outer-membrane proteins as determined by sodium dodecyl sulfatepolyacrylamide gel electrophoresis [1]. This subtyping method has proven useful in epidemiologic studies and was recently expanded to include 21 different subtypes [3]. Kilian has described a different method for the subclassification of Haemophilus species, which is based on biochemically determined biotypes [2]. In the present study we compared the two subclassification systems. Biotype I accounted for 89% of the 256 isolates tested. Biotypes III and V were not observed. Analysis of outer-membrane proteins permitted biotype-I strains to be subdivided into several different categories. Subtype IH was the most common (44070), and most of the remaining biotype-I strains had other common subtypes. Biotype-Il and-IV strains were observed almost exclusively among subtype-3L strains or among strains with rarely encountered subtypes. The difference in the distribution of subtypes among biotype-I strains and strains other than biotype I was highly significant when isolates were grouped by subtype into the following categories:(1) common subtypes other than 3L,(2) subtype 3L, and (3) uncommon subtypes (X2= 79.1; two degrees of freedom; P< 10'8).
