Peptide-chain elongation using unprotected amino acids with mixed carbonic anhydride was achieved. Micro-flow technology enabled the rapid mixing of an organic layer, containing a protected peptide or a protected dipeptide, and an aqueous layer, containing an unprotected amino acid or an unprotected dipeptide to accelerate the desired amidation, which successfully suppressed the undesired racemization/epimerization.
