TIlE COMPLEMENT SYSTEM is composed of a number of serum proteins that, when functioning in an orderly and integrated fashion, serve as important mediators of host defense and inflammation? Activation of the complement system can either be beneficial to the host, as in resistance to invading microorganisms, or it can be detrimental to the host, as in a variety of immunopathologically mediated disease.During the past two decades much new knowledge concerning the genetics of the complement system has led to a better understanding of several clinically significant conditions. We summarize current knowledge about the complement system, the genetic basis for its variability among individuals, and the relationship of that variability to human disease.
