CHROMOSOME INSTABILITY IS ASSOCIATED WITH HYPODIPLOID CLONES IN MYELODYSPLASTIC SYNDROMES

Chromosome study on 30 patients with a myelodysplastic or preleukemic syndrome was performed. Fourteen patients had an increased number of cells with chromosome instability, e.g., non-clonal chromosome aberrations. The patients in the instability group had more clonal chromosome aberrations (12 out of 14 patients) than did patients in the non-instability group (7 out of 16 patients). In the instability group 5 patients had a hypodiploid clone and 8 patients had a clone with an aberration in chromosome 7, whereas these aberrations were not seen in the non-instability group. Chromosome instability associates strongly with hypodiploid clones and/or with aberrations in chromosome 7. Chromosome instability associates with previous chemotherapy and/or radiation therapy (only 1 out of 7 patients with non-therapy belonged to the non-instability group). Half of the patients in the instability groups had no previous chemo- or radiation therapy. The causes of chromosome instability in these patients are discussed.
