Materials and methods Sources and isolation of fungi from diseased plants. Canola plants with root rot-like symptoms were collected in 1984 between 27 June and 4 July (seedlings) and between 27 July and 2 August (adult plants) from 70 fields in the main canola growing areas of central Saskatchewan. Roots and basal stem portions with distinct lesions were cut in pieces, rinsed in running tap water to remove soil, surface sterilized with 10% Javex (0.6% NaOCl)(1 min seedlings and 2 min adult plants), and plated (4 pieces per petri dish) on potato dextrose agar (PDA) supplemented with 0.001% streptomycin sulphate. Two to 30 root/stem pieces from each sample were used. Plates were incubated at 20 C and examined every 24 h for one week. The frequency of occurrence of different fungal genera was recorded but no attempt was made to identify species.
