Microiontophoretic administration of acetylcholine or 3′,5′-cyclic guanosine monophosphate powerfully excites pyramidal neurons in hippocampal formation transplanted to the anterior chamber of the eye. Responses to both these agents are antagonized by atropine and potentiated by the phosphodiesterase inhibitors IBMX and RO 20-1724. Acetylcholinesterase-positive fibers, derived from the cholinergic input to the iris, invade the transplant and ramify diffusely within the brain substance. After ablation of the ciliary ganglion, such fibers are not seen in the iris or transplant. Excitatory responses to acetylcholine and cyclic GMP are readily seen, however, after both ciliary and superior cervical ganglionectomy. These experiments demonstrate that cholinoceptivity and cholinergic afferent fiber input, characteristic of normal hippocampus in situ, are retained in the transplant and suggest that cyclic GMP may be involved in cholinergic transmission in the mammalian central nervous system.
