Materials and Methods STRAIN.-Trichoderma harzianum (Rifai2l) OMZ 779 was isolated from a piece of decomposing bark found in a forest near Brugg, Switzerland.POLYSACCHARIDES.-Mutan, the insoluble extracellular glucan from Streptococcus mutans OMZ 176, was synthesized from su-crose by a mixture of glucosyltransferases, as reported previously.'9 Modified mutan was prepared from native mutan by periodate oxidation and reduction.'l The insoluble gellike dextran from Streptococcus sanguis 804 produced by a mixture of glucosyltransferases was a gift.* Nigerose, nigerotetraose, pachyman, sclerotium rolfsii gum, pseudonigeran, laminarin, and nigeran also were donated. t Fractions from dental plaque containing insoluble matrix polysaccharides were prepared according to the methods described elsewhere. 22 REAGENTs.-Sephadex G-150 and blue dextran, t Ampholine, § pH 5-8, and Aquacid IIII were purchased. All other chemicals were reagent grade, and carbohydrates used were of the D configuration. CULTURE METHODS.-Trichoderma harzianum OMZ 779 was maintained on malt agar.# For enzyme production, this strain was grown in a basal medium meeting the
