T HE RATIONAL DESIGN of strategies to avert rejec-tion of xenografts requires further characterization of the immunologic response. It has been shown in experimental systems that the cellular mechanisms responsible for the host response to xenografts differs from that of allografts, being more dependent on the following:(1) CD4+ T cells,’(2) the indirect pathway of antigen presentation,’and (3) preformed or induced humoral responses.’With these principles in mind, we postulated that an ideal xenogeneic recipient would be deficient in CD4+ T cells and incapable of both indirect presentation and the production of specific xenoantibodies. To produce such recipients, we cross-bred major histocompatibility complex (MHC) class II deficient (CIID) mice with mice lacking B lymphocytes (p--I-) due to a disruption of the immunoglobulin M heavy chain gene. These mice which lacked CD4.’T cells and B cells (CIID xp-/-) were tested for their ability to reject allogeneic and xenogeneic pancreatic islet grafts.
