The expression of theompFandompCgenes that code for major outer-membrane proteins ofEscherichia coliis positively regulated by the products of theompRandenvZgenes. Recently, we isolated theompR77mutation, which suppresses theenvZ11mutation. In this work, a novel mutation that interferes with the suppression of theenvZ11mutation byompR77was isolated. The mutation was located in therpoAgene that codes for the α subunit of DNA-dependent RNA polymerase. These results suggest that an interaction between the positive regulators and RNA polymerase is involved in the initiation of transcription of theompFandompCgenes. In addition, the results suggest that during transcription the RNA polymerase migrates along DNA strands with the α subunit facing backward and the ββ′ subunits facing forward.
