METHODSMice. All mice were purchased from Jackson Laboratories (Maine) or bred in the facilities of National Jewish Center (Colorado).T-ceU hybrids. The T-cell hybrids used in this study have been described previously (Bill et al. 1988). They were generated from mitogen (concanavalin-A)-stimulated peripheral T cells from B10, B10. Q, and B10. BR mice that were fused to BW5147 and maintained in culture as described previously (Kappler et al. 1981). Selected for use in this study were 24 hybrids that expressed mRNA for Va5. Sequencing the/3-chain eDNA from the T-cell hybridoma 2HB51 showed that this hybrid expresses an in-frame V, 5.1 mRNA transcript. 2HB51. 8 is a subclone of 2HB51 selected for high levels of Va5 RNA.
