Proto-oncogene fos is expressed during pre-and postnatal development of the mouse, cellular differentiation, and cell proliferation. Day-17 to-18 mouse amnion displays high levels of both fos-specific 2.2-kb transcripts as well as 55K los protein. Bone marrow cells and macrophage cell lines express fos transcripts. Rapid induction of fos gene expression is witnessed when either a human promonocytic cell line, U937, or monomyelocytic cell line, HL-60, is treated with phorbol ester (TPA) to differentiate to mature macrophages. The highest levels of induction occur by 30 minutes but fos RNA is synthesized continuously, albeit three-to fourfold lower, for 96-108 hours. In contrast, fos protein can be detected for only 90-120 minutes postinduction. When PCI2 cells differentiate to form neurites upon addition of nerve growth factor (NGF), thefos gene is rapidly but transiently induced, with maximal levels accumulating by 20-30 minutes. Unlike the monocytic cell differentiation, no fos-spe-cific transcripts could be observed after 120-240 minutes of addition of NGE Rapid and transient induction of the expression of thefos gene is also observed when quiescent mouse fibroblasts are stimulated to proliferate with mitogens. In an attempt to understand the regulation of transcription of the fos gene, a transcriptional enhancer element has been localized to reside between-60 to-400 nucleotides upstream of the 5'cap nucleotide. We discuss the complex regulation of thefos gene in terms of its role in normal cells and its ability to induce cellular transformation in vitro.
