ANTIBODIES TO EPSTEIN-BARR VIRUS-DETERMINED ANTIGENS IN NORMAL SUBJECTS AND IN PATIENTS WITH SEROPOSITIVE RHEUMATOID-ARTHRITIS

Patients with seropositive rheumatoid arthritis (RA) have an increased frequency of precipitating antibody [Ab] against a nuclear antigen [Ag], the RA nuclear Ag, detected in human B [bone marrow-derived] lymphoblastoid cell lines infected by Epstein-Barr virus [EBV]. Patients with seropositive RA have specifically elevated titers of Ab to another, better-characterized EBV-associated B cell Ag, the EBV nuclear Ag, which is detected by anti-complement immunofluorescence. Titers of these 2 Ab were not affected by absorption of rheumatoid factor from serum. Patients with RA did not have elevated titers or Ab against the EBV capsid Ag or to 3 other species of human herpesviruses: herpes simplex type 1, varicella-zoster virus and cytomegalovirus. In normal individuals and RA patients there was a significant association between the presence of Ab to RA nuclear Ag and the titers of Ab to EBV nuclear Ag. Normal subjects with Ab to RA nuclear Ag had titers of Ab to EBV nuclear Ag equivalent to those of patients with RA and significantly higher than normal subjects lacking Ab to RA nuclear Ag. Patients with sero-positive RA may derive from a larger population with enhanced immune responsiveness to B lymphocyte nuclear Ag determined by EBV.
