Tumors of Schwann cell origin represent the most common clinically important manifestation of neurofibromatosis (NF).'.'Prior studies have noted that neurofibromas may develop or enlarge during puberty and pregnancy and that acoustic neuromas and neurofibromas contain cytoplasmic and nuclear binders for sex steroid horm0nes. 3'~ We postulated that the growth of Schwann cell tumors could be modulated by hormonal rnanip~ lation.~ Since no naturally occurring animal model for NF is presently available, we studied the effects of castration, 17-6-estradiol (Ez), and the antiestrogen Tamoxifen on the onset and growth of Schwann cell tumors induced in rats by parental administration of N-methyl-N-nitrosourea (MNU).
