LEUKOKINETIC STUDIES .4. TOTAL BLOOD, CIRCULATING AND MARGINAL GRANULOCYTE POOLS AND GRANULOCYTE TURNOVER RATE IN NORMAL SUBJECTS

The size of the total blood granulocyte pool (TBGP) and its two subcompartments, the circulating granulocyte pool (CGP) and marginal granulocyte pool (MGP) have been measured in 70 normal male subjects. In addition, the half-time disappearance (T 1/2) of granulocytes from the blood and the daily granulocyte turnover rate (GTR) were determined in 45 of these subjects. The mean values [plus or minus] 1 SD, expressed as number of cells X 107 per kg of body weight were TBGP, 65.0 [plus or minus] 22.4; CGP, 31.7 [plus or minus] 11.1; MGP, 33.3 [plus or minus] 16.0. The mean T 1/2 was 6.6 [plus or minus] 1.16 hours and the GTR was 179.9 [plus or minus] 74.3 X 107 granulocytes per kg of body weight per day. Acute physical exercise and epinephrine infusion produced a shift of cells from the MGP to the CGP but did not change the size of the TBGP nor alter the GTR. Oral prednisone given for 11 days resulted in an increase in size of the TBGP with both the MGP and CGP increasing proportionally. At this new blood granulocyte level a steady state was established with a normal GTR. Intravenous bacterial endotoxin injection produced a shift of cells from the CGP to the MGP and an influx of cells from the bone marrow granulocyte pool resulted in an increase in the size of the TBGP.
