REQUIREMENT OF ENERGY FOR CELL FUSION REACTION OF EHRLICH ASCITES TUMOR CELLS BY HVJ

The cell fusion capacity of Ehrlich ascites tumor cells with HVJ was strikingly increased by preincubating the cells under aerobic conditions at 37[degree]C, while it was decreased by preincubating them under anaerobic conditions, and this change in the cell fusion capacity was reversed by reversing the preincubation conditions. The change was not due to differences in the quantity of virus particles adsorbed onto the cell surface but was related to the content of nucleotide triphosphate (NTP) in the cells. Puromycin and actinomycin did not inhibit the change in the cell fusion capacity on preincubation or the cell fusion reaction itself. From studies on the effect of adding 2,4-dinitrophenol to inhibit oxidative phosphorylation during the course of the cell fusion reaction, it was concluded that the following reaction steps occurred in the cell fusion reaction virus adsorption onto cells and agglutination of these cells, disconnection of the cell surface by the action of HVJ at 37[degree], reconnection of the disconnected sites or their fusion with those of neighboring cells, a step requiring NTP, and spherical giant cell formation.
