A 24-HOUR PERIODICITY IN THE LH-RELEASE APPARATUS OF FEMALE RATS, DISCLOSED BY BARBITURATE SEDATION

Earlier investigation by the use of Dibenamine and atropine indicated that in 4-day cyclic rats in the authors'' colony, a specific neuro-genic stimulus, essential for the ovulatory discharge of LH, activates the hypophysis between 2 and 4 P.M. on the day of proestrus. Evidence of a 24-hr. rhythm of excitability in the neural component of the LH-release apparatus was also recently reported. The present results confirm the earlier ones by different methods. Nembutal (pentobarbital Na[long dash]standard dose 30 mg./kg., intraperit.) prevents the ovulatory activation of the hypophysis when administered at 2 P.M. during proestrus. Persistence of graafian follicles for 2-3 days results when Nembutal treatment is repeated on successive afternoons at the same critical hours. On the 2nd and 3rd days a supplementary injn. is required at 3:30-4 P.M. If, on any of these 3 days, treatment is omitted or postponed until after 4 P.M., pituitary activation will occur promptly and ovulation will follow during that night. Similar results are produced by amytal, dial, barbital and phenobarbital. Blockade of LH release by treatment with the barbiturates on successive days discloses a follicular cycle which terminates in atresia about 2 days before onset of the next proestrus.. Consideration is given to similarities and differences between such follicular cycles and the overlapping ones found in "spontaneous persistent estrus" and "light estrus." The 24-hr. rhythm, although possibly a property of the LH-release mechanism in other spontaneously ovulating forms as well as the rat, cannot be considered as a universal characteristic until a variety of animals has been investigated.
