The cyclodextrins (CD) are a homologous series ofnon- reducing, cyclic, D-glucose polymers with six or more glycosyls in a ring structure.1} These dextrins are synthe-sized from starch and related materials by cyclomaltodex- trin glucanotransferase (CGTase, EC 2.4.1.19). These enzymes are produced by several microorganisms.2~6) Alkalophilic Bacillus sp. strain No. 38-2 produces an extracellular CGTase which led the cyclodextrin industries. We have cloned the CGTase gene of alkalophilic Bacillus sp. strain No. 38-2 in Escherichia coli with pBR322to enzymatic The chromosomal DNAof strain was partially cleaved with Sau3AI and the fragments were ligated to BamHl digested pBR322. A transformant carrying the plasmid the has the CGTase activity.
