PACAP is a new member of the vasoactive intestinal polypeptide (VIP) family of pep-tides1 present in the brain2–7 as well as in neuronal elements of a number of peripheral organs. 8–12 PACAP is expressed in nerve cell bodies of the dorsal root ganglia, 13–14 trigeminal ganglion, 15 and mesencephalic trigeminal nucleus, 16 suggesting a sensory role for the peptide. In the present study we have determined the contribution of PACAP sensory nerve fibers to the innervation of rat visceral organs by (1) examining the effect of neonatal treatment with the sensory neurotoxin capsaicin on PACAP immunoreactivity, determined by radioimmunoassay and immunohistochemistry, and (2) using double-immunostaining for PACAP and calcitonin gene–related peptide (CGRP), a peptide known to be stored in capsaicin-sensitive sensory nerves.
