THE umbilical circulation is the vital link between the mother and the fetus in its intrauterine existence; not only is it the pathway for gaseous and metabolic exchange, but it is also the single most important determinant of systemic vascular resistance within the fetal circulation (Dawes, 1962). Biological vasoactive substances which stimulate cord and placental vessels are therefore of interest in fetal physiology and pharmacology, and these have been the subject of much investigation (Euler, 1938; Panigel, 1962; Takaneka, 1963; Somlyo et al., 1965; Karim, 1966, 1967; Astrom and Samelius, 1957). At birth the most important function of the cord vessels, especially in animal obstetrics, is complete closure, and an adequate physiological mechanism appears to exist for the cessation of umbilical blood flow (Rachmanow, 1914; Price, 1944; Williams, 1931). Karim (1966) considering the possible involvement of a chemical substance in closure of the cord vessels at birth investigated extracts of cords from spontaneous vaginal deliveries at term, and found several smooth-muscle-stimulating hydroxy fatty acids. These were later identified as prostaglandins El, E,, F,, and F,, (Karim, 1967). These prostaglandins were shown to be active or: the longitudinal strips of the umbilical artery. PGE,, PGE,,, PGF,, were found to cause shortening of the isolated muscle and PGE, relaxation. An investigation into the effects of these four prostaglandins on isolated circular and longitudinal muscle preparations of human umbilical and placental vessels at term and at various stages of pregnancy has been carried out.
