2 MESSENGER-RNAS CAN BE PRODUCED FROM A SINGLE IMMUNOGLOBULIN-MU GENE BY ALTERNATIVE RNA PROCESSING PATHWAYS

The .mu. chains of the membrane-bound (.mu.m) and secreted (.mu.s) forms of Ig[immunoglobulin] M are encoded by 2 species of mRNA. Cloned c[complementary]DNA produced from the 2 .mu. mRNA of M104E mouse myeloma tumors differ only at their 3'' ends, which encode either the .mu.m or .mu.s C terminus. The .mu.m and .mu.s mRNA are produced from transcripts of a single .mu. gene. The last 187 nucleotides of .mu.s mRNA are derived from DNA contiguous with the 3'' end of the sequence encoding the C.mu.4 domain. The .mu.m cDNA clone does not include these 187 nucleotides, but instead contains 392 nucleotides derived from 2 exons located 1850 bp 3'' to the C.mu.4 sequence. Comparison of genomic and cDNA sequences show that in .mu.m mRNA, an RNA splice of 1850 nucleotides joins a site in the coding sequence at the end of C.mu.4 with a site at the beginning of the 1st membrane-specific exon. A 2nd RNA splice of 118 nucleotides joins sequences transcribed from the 1st and 2nd membrane-specific exons. The differences observed between .mu.m and .mu.s cDNA suggest that developmental control of the site at which poly(A) is added to transcripts of the .mu. gene determines the relative levels of .mu.m or .mu.s chain synthesis. Possible models for the control of .mu. gene transcripts and the significance of this form of developmentally regulated RNA processing for the evolution of eukaryotic split genes were discussed.
