In order to minimize the effect of flashlamp instabilities in determining fluorescence decay profiles by the time-correlated single-photon counting technique, an alternate recording of excitation and emission profiles is advisable. In the equipment described here, this is performed by changing the light path via two quartz prisms mounted on a horizontal plate and moved into alternate positions, while the sample remains fixed. This design is extremely simple compared to other methods reported, since the sample holder is left unchanged. The efficiency of the equipment was tested by the analysis of double exponential decay curves using fluid solutions containing anthracene and di-meso-substituted anthracenes, which have almost temperature-independent and strongly temperature-dependent lifetimes respectively.
