This paper is concerned with the five polypeptide chains that constitute the nicotinic acetylcholine receptor (AChR). We have used electron microscopy to locate the chains relative to one another. We have also used affinity labels and time-resolved photolabeling to identify functional sites on the chains other than the ACh-binding sites. The background for these studies has been reviewed by Karlin (1980), Changeux (1981), and Conti-Tronconi and Raftery (1982). Our conclusions are (1) that the five chains of the receptor are arranged around the central axis in the order a3, a~ 5 and (2) that the a-and B-chains contribute to functionally significant local anesthetic binding sites.
