METHODSThe sheep used in this study were all purebred Dorsets. Fetal blood was obtained from the umbilical vein in the last third of pregnancy. Adult sheep blood was obtained from nonpregnant ewes. Blood samples for Oz-dissociation curves and red cell organic phosphate concentrations were obtained in the first 2 months of extrauterine life. Oxygen-dissociation curves. The oxygen-dissociation curves were constructed as described previously (7). pH and Paz were measured at 39.6 C. The POT was measured with a Clark-type electrode connected to a Beckman gas analyzer, model 160. Oxygen tension determinations on a standard gas mixture, with a Pea in the same range as the blood samples, were run before and after each PO:! measurement on a blood sample. pH was measured with a Radiometer model 4 pH meter and a type E 5020 microelectrode chain. In fetal and adult blood, oxygen saturations were measured spectrophotometrically with a Radiometer oximeter on hemolyzed packed red cells. The oximeter was calibrated each day by determining oxygen contents and capacities on selected blood samples with a Beckman GC-2A gas chromatograph. Since blood obtained from lambs postnatally may contain a more heterogeneous population of red cells in terms of their oxygen affinities, the 02 saturations of these blood samples were calculated from the determination of oxygen contents and capacities by gas chromatography to avoid any errors introduced by the use of spectrophotometric techniques. The results of the oxygen-dissociation curve were expressed in the form of a general equation:
